Wiring up the sherline cnc mill to the controller board

After sitting in various garages around town for the last two years it's finally moving forwards... many thanks to the emc IRC chanel on freenode.

A breakthrough in my understanding and general confidence came at the hands of Andy Pugh who walked me through the laypersons continuity test

the motors are rated 3.0V, 2A, 1.8 degrees step on the xy axis and 4.5V, 1.4A, and 2 degrees step on the z axis.

I've got a 24V 1A DC power supply handy, as well as a 12V 1A that perhaps I may wire together to get a bit more juice.... still gotta figure that stuff up :)

OK, this will also work: touch motor wires together in pairs and find which pairs make the motor harder to turn.

   andypugh:   It shows that you can use all or half of the coils, by connecting to each end, or to one end and the middle.
   andypugh:   There is another sort of stepper driver where you connect the centre of each coil to earth/ground and then apply the power to one end or the other.
   andypugh:   That is called "Unipolar"
   andypugh:   But you have a bipolar driver, which effectively swaps power and earth as required.
   andypugh:   It doesn't really matter which end of which phase (A or B) you connect to the driver, as long as you make totally sure that you don't connect one phase to another. (I did that, blew my driver).
   andypugh:   ie, make sure that of the two wire families you have seperated, that one set goes to the "A" terminals, and one to "B"

anyhow, with thtat cleared up the three axis are now wired... it's time to link up the power cords :)

blender fluid text in 2.49 plus some cool inspeck stuff

I'll lay off the blender tutorials after they seem better answererd on the net.... until then... blender fluid text is really fun and cool, however getting it to work correctly is a task that's been nagging me and my student at http://computers4kids.net for the last two weeks.... well the first real decent news is that in response to several threads out there you don't need to up the bake resolution to 300 (requiring several gigs of ram) you MAY however need to enter every letter as an individual mesh.... it's a chore, however the results are great...

The leaf in the picture is actually an inspeck 3d pic of a piece of lost styrofoam sculpture made a few weeks ago... am very pleased with the outcome, however there seems to be some artifact in there somewhere....

hope to update this video once the artifacts are gone (and once I figure out how to get the alpha chanel ipo's a little nicer....

kept walkin down the tracks and stumbled upon some excellent green foam and very fine sand - knew it was gonna be a blingin day.

paid a visit to the job site to process some blingless rims that have some bangin aluminum...

if the small track hoe doesn't work...

The model as sent to the CNC machine - the beading is there, however some of the detail is missing... at 1/4 scale this translates in to less than a 64th of an inch... it should be ok

The doorway in miniature waiting for the freeze and doors to be mounted. The miniature hinges designed to open both doors simultaneously were created by hand here in the valley. These doors were designed by Thomas Jefferson and are in use presently at Monticello

The painted 1/4 scale model - notice the hand carved beading on the angle - another local artisan recreated this. Great attention to detail - it matches the horizontal beading, however includes the ~33 degree angle.

In perhaps a giant effort to get a great pic of Trevor - one of Gaston & Wyatt's de facto mascots - the Inspeck 3D digitization scannerfinally got cleaned up and put to work... The first pic shows the original image, the second shows the detail in 3d... probably need to put some polish on this, it's a start though... In the first picture (taken from a 3/4 scale model made by a local artist...) the beading around the bison's horns can be seen in clear detail - there's definitely some work to be done in the conversion if a full scale model is going to be usable, however for reducing to 1/4 scale (ie the final model is about 1" in diameter... ) and further having a layer of paint applied.... these models are quite sufficient. As they are carved in wood and MDF board the finest expected resolution should be in the order of 1/16 - 1/32 of an inch. In aluminum the results may be superior...

All told almost a dozen people came together to make this project a reality - here in no particular order is credit where due

Viva le lost foam technique

After almost three months of playing with the silly stuff at work it finally dawned to test the lost foam technique - massive success. To the left are homey's initials carved in blue foam. To the right are those initials in aluminum. Absolutely wicked.
Lessons learned: Don't pack the sand too hard or air bubbles get in there. Too loose and the metal may pour out the side of the foam (especially when you pack it up around the edges of the design... This piece came out great and the little dude tore outta there 30 seconds after completion to show his bros. Most excellent.

DESMOND Part 1

I am behind the times so often it is startling. Dr. Kretsinger took some time on a recent hike to up it to me bluntly "It would help if you caught up to physics 1908." With that in the background, attendance at the Chem E. lecture - arranged by the Shirts Group - made for an enjoyable blitz-crash.

One of my favorite facets of America is the influx of great minds generated of great cultures. Kolossvary is of Hungarian descent, and very much looks the part - broad shoulders, thin mustache, and Mediterranean complexion... with one notable exception... the fingers of a programmer. Much like a pianist, a programmers fingers, albeit strong, are often long and dextrose. In any case Dr. Istvan fits the part of the guy who has sledge-hammered on a chemo-physio-computer engineering project around for the past 17 years. My guess is this guy's keyboards last ~3-5 months max.

At first blush DESMOND has some impressive stats. Dr. Shirts of folding@home fame chuckled throughout the lecture. At this slide he cited the first microsecond of modeling required 6 months to calculate.
Towards the bottom of the slide note the Anton - this processor, also developed by D. E. Shaw Research, works in tandem with Desmond to crunch up to 14 microseconds per day.

To facilitate we amateurs Desmond comes with a free downloadable Desmond GUI merged with Schrodinger's Maestro software. An impressive and thorough package a full license for Maestro runs in the $30K+ realm. The academic license, however, is free and provides an excellent platform to enjoy Desmond from. The pros: In fifteen minutes or so I had gone through some basic protein preparation wizard and was running Desmond through operations and getting a bi-lipid membrane like the pictured above. Amazing stuff that might have taken every bit of a month a few years ago to understand and initiate now accomplished in minutes.

Beneath the GUI hood there are even more features. To access these Desmond also comes with a command line interface - this provides additional algorithms for more in depth view and control. Navigating between the gui and command line is helped greatly by Maestro's process and file management that creates files prior to executing actions that are very simple to read and modify. Shirt enthusiastically likened these unto a voting paper trail, and I must agree it's a handy feature.

Cons: well it's not too big a deal, however there's no 64 bit binary yet, and for simplicities sake I ended up running Maestro in a virtual 32 bit machine to get everything more stable. Dr. Kolossvary indicated that this upgrade might be coming as early as this month - so stay tuned. Another planned feature includes direct reading of OPM's protein database files.

This week is finally calming down and am going to start running some simulations - stay tuned for part 2 some time in the next week or so.

The Making of Membranes

After importing a PDB file go to the protein preparation wizard under "workflows" on the far right hand side
Dr. Kosovarry recommended assigning bond orders, adding hydrogens, treating metals, capping the termini, and detecting the disulfide bonds. In his example he did not feel it necessary to delete waters... there are some other options below that are only available with the full version.... ah well... it's a start. Also there appear to be some missing AA groups and I am yet to figure out how to fix that...
Now under "Applications" go to "Desmond" and go to the System Builder. There are 3 solvent models available, with more on the command line - There's an ion placement submenu perhaps a ligand placement menu may appear in future versions.
The membrane builder menu let's you autoposition the membrane using selected atoms or amino acids - I've had moderate success in this using the selection tool to grab a bunch of atoms around the disc created by the OPM database, If you're not satisfied with the initial placement modify it clicking the "Adjust..." checkbox.
When you're ready to see your model click "Start" - depending on a 2cpu 3.2 ghz machine you'll see a model like the above in a minute or two with ~120k atoms in the case of FAAH.

That's right- he's our Mexican Jesus... but seriously ya'll as our nation debates as to "what to do with all these immigrants" I would encourage the following line of reasoning these "Mexicans" are vastly more "American" than your average honky walking on the street. Their ancestors were here 25,000+ years before most of the people reading this got here. Jesus here speaks some of the original Aztec language... how many "Americans" speak ONE WORD of native tongue. Most "Americans" have been in the USA less than 100 years, and are now trying to perform a repeat of the conquistadors actions. Viva la revolucion, and to all you "Mexicans" - please move to Charlottesville. We need more of you. Ya'll are way cooler than most of these immigrants from the North.

Well thanks to VMD, Pubchem + Pubchem Edit + Blender 3d + Biocyc.orghere's a test video of a hypothesized cannabigerolic acid synthase (aka the geranyl diphosphate transferase) that converts olivetolic acid (painted in blue + gray) into cannabigerolic acid (painted with red) need to get the gpp in there correctly and for those who must know the protein is 1U0V - I picked it for a couple reasons, however it's sorta silly cause it hasn't been structured. It's for illustration.

Charlottesville Community Bikes, Aluminum Foundries, and Bling

Had a rad time at the Cville community bike shop this weekend. We melted a bunch of aluminum and one lucky gal got some handmade hard core bling for her wheelz. And every one got to hear a bunch about aluminum, aluminum oxide, steel, slag, and a bunch of other stuff that makes the whole thing so what it is... rad.

getting vrml / wavefront from molecular data into blender 3d . vol 1

So far getting the VRML output from different softwares has been a bit of a chore. We started out using

Rasmol. It renders the sticks in VRML however it doesn't do the ribbons.... stay tuned for updates this summer - the developers have noted the bug and emailed me back to say they're aimin' to squash it
Chimera It renders the VRML for anything, however having a time getting the files in to blender
VMD Halelujah - well it's a different approach altogether... and who'da thunk. Had to use an older version to run it on my osx 3.xx powerpc and... score - rendered to wavefront and... shazaam...

Well least surprising surprise d'jour - PDB2Blend is some wiley kraut's python script for getting the data straight - doesn't do everything VMD does... ball and stick, space fill, and a stick.... still very cool

This was the 17th day of the campaign.

At 0020 hours the radio operator for headquarters listened to the message tapped out, grinned and then handed it to a runner. The runner looked at the message, also smiled, and then took it to the Staff, still up and about in the small black operations tent. Captain Dreier looked at the message, nodded his thanks to the runner and dismissed him. Next to get the news was the Colonel, who announced the news to all the officers.

The 4th Tabor would be attached to the 1st Infantry Division, and would operate on the left flank of the Division, wearing French helmets and American fatigues.

Translated, that meant a regiment of Moroccans called Goums, composed of 4 companies, would augment the division effort. These Moroccan warriors with their pigtails and brown burnous’ were not entirely new to us, either in this war or World War I. In the great summer offensives launched by the Allies in 1918, the 1st Division had operated with the First Moroccan Division on its flank. And in this war, during the Ousseltia Valley campaign, these deadly raiders had spread death and terror among the German and Italian units on the djebela guarding the road to Kairouan. The Axis troops knew that these Goumiers had a reputation for not taking any prisoners, and for delighting in fighting at very close quarters – preferable with knives. Now they were to work with us, and the chances were that the Italians and Germans would know it double-quick time the Goumiers were around. No wonder the officers grinned. They knew the short fierce raids that the Goumiers liked to launch. And they knew the incessant guard duty it entailed to ward off such sudden blows; it was bound to affect morale.

Let me spare you the details to start out with (besides, everything listed alphabetically puts editgrid in first anyhow)

editgrid.com: comin' at ya live from Hong Kong, this bad boy has it all. There's really not a good reason in my mind to review the other commercial piece (jot.com) except to trash it and save you the reader some troubles. EditGrid is amazing, uploaded an xls file with a couple thousand rows. Formatting - retained! Multiple tables in a spreadsheet? No Problem! EditGrid has it all. This is a straight up killer ap. Posted the entire city of Charlottesville line-item budget and voila! Up in a few minutes. Not surprisingly this cite is getting hit hard at the mooment, expect some delays while their netadmins deal with the reality of being excellent.

Jot.com (now google's love child, and prolly soon to move towards non-suck is a nice application that bites bytes). It's being revamped right now, this "lucky" author got to check it out pre-google, so there's no telling what the new release will do, check back in a few months, it's google, and it's bound to get better with some resources. So far however.... nothin' werked, as they say. Cut'n'Paste your spreadsheet and watch your computer crash and burn. No format retention and formulas don't work. This thing sucks

wikicalc : Dan Brinklin worked his way in to the geek hall of fame with visicalc almost 20 years ago. His new spreadsheet wikicalc is open source gpl, and so it gets some big points for that. Indeed with a decent server this may be a killer ap. It also has some decent cut'n'paste features, however it flakes out pretty hard on a serious data load. On the plus side it retains some formulas and can do formatting - WAY BETTER THAN JOT.COM and free open source! Even its alpha release is more inspiring than the jot.com crud.

wikicalc, jotspot, charlottesville city budget

Wikicalc deserves some props for being a groovy open source spreadsheet wiki that's very decent for it's price, and works through some crucial bits of excel, so as to retain basic formulas and such.

With jot.com going to google.com last week there's another bunch of kids in the same playgroundsure to bring good attention to the arena where more transparent business practices can be developed, standardized, and optimized. Their product is sleek AJAX based and with calendar and location parsing technology built in this is one another slick development for the google folks.

And so that brings it around to the c-village 'hood skool. - charlottesville.jot.com - starting out with some of the non-general stuff, and working towards the line item morsels.

Carnegie Mellon Technology in the Arts

had a great time. Greeted at the entrance of CMU by a talking computer that seemed to recognize my lostness - "Do you need directions" came the metallic max headromesque voice.... I wasn't sure how advanced this computer was and assumed it was referring to direction in the sense of a few meters, as opposed to some greater direction in life....Anyhow, i responded yes about the time a friendly human concierge arrived to get me to the first meeting of the day. Not sure whether the human was controlled by the computer or whether it was of higher elegance... anyhow....

And as it happened the first meeting "Funding Technology Projects" was right down the hall. Met with Mr. Mackie of the Andrew Mellon Foundation - he is the head of Research in Technology Funding. He emphasized open source "if you're looking to spend on proprietary software look elsewhere" (http://www.mellon.org/research_it.html) and also emphasized use of open standards (The Mellon Foundation (along with MIT and others) funded dSpace, which is something we already ascribe to through Greenstone). **** If we're interested in collaborating on a school bus full of digitization equipment this would be the guy to talk to. they are playing with huge sums of money and we should really try to start something with them "if we fund you once we'll probably fund you again" -- in other words they like long term relationships.

Although initially slated to speak on Joomla's content management system (marshallrotc.org Virginia Verified Voting) when I got to looking at the schedule two other presenters wanted to cover Joomla and so I shifted my focus to Greenstone and IBM's analysis of wikipedia showing the underlying stability of the wiki format. Also met with Ellen Arnold, who invited me to be a grant reviewer for the Institute of Museum and Library Services (IMLS). I suspect this is a bit like being invited into the book of the month club, anyhow, I was earnestly flattered and accepted.

Otherwise enjoyed Pittsburgh a fair bit. Solid steel town, and affordable housing - imagine paying under 70K for a *solid* brick house? Went up Washington hill for the view, ate a hoagie, and went to a comic book convention across the street from my lodging. Very glad to be back, however must admit Pittsburgh is a most impressive and down to earth city. I also studied some of the requirements for funding in Pennsylvania, and would be glad to pass on some of that to the folks in Carlisle if they're not already well aware. The Bayer Foundation seemed very interested when I was talking about library->library services.

OK - started this a few months ago, and really not totally convinced Koha is gonna do what I need - neverthless it's worth a look if all you've got are a bunch of Marc Records. Greenstone.org's product, however, handles marc and a gajillion other formats....

The Enhanced Koha LibLime style is an amazing piece of code, picking up where the left of the geeksquads in dev land leave off - integrating a functional and user-friendly Amazon.com row into the matrix let's users "buy it now" from amazon while browsing through the collection in Koha gives libraries looking to justify digitizing their collections added incentive.

Getting Koha running can be a bit of work, depending on your operating system. IndexData does most of their work on Debian based systems (think Ubuntu) - and if you're running anything else it may take a few tries to get the whole thing running.

For a better understanding of the install process they have created a wiki and decent documentation.

Well it's been something I've been looking for forf a long long time - Webhuddle finally provides an open source VOIP + Desktop Sharing + Whiteboard software tool for the open source community. the developers are cool, and the forum is starting to take off. if this keeps up most websites will wiki+whiteboard their way through development processes plus database meetings and discussions for training. VRML Bridges, powerlines, buildings, tanks, aircraft, helicopters etc... totally badass - all in VRML 1.0 specification.

To convert these guys for white_dune (VRML 2 (some folks call it VRML97) you can use a nifty piece of software called Crossroads 3D if that's not doing it for you (it can drop out occasionally on large files) the vrml1 to vrml2 converter is a nice commandline utility that whoops.

Once you've got everything in VRML 2 (VRML 97) you can begin working in white_dune - the biggest challenge their is the preview window and moving objects around in there - if you've got a 3d mouse / joystick you may have more success and simplify your life - otherwise it's not bad.

lightscribe is the most awesome technology i have witnessed - amazing stuff. professional quality out of the box badsas anyhow, it's rad. on average a full image takes anywhere from 15-25 minutes to burn - the time is well spent - the design package that came with the pack "prints" the design, the package prompts you all sortsa blah blah. it's worth answerin because after all - blah blah reminds you that some bastard over yalls onder and back had to program that blah blah and it said a load of jack that actureally mattered. programmers all over the planet right now are laughing about this that and the other php perl myscript or your script or whatever have you - and props out to for givin me a such an awesome technology as a dual-purpose laser etching through microns of material... i'm sure someone will come along and try to make the whole thing faster, believe me though -buyer beware - the time frame is awesome on this model - between every print a pause - time to reflect on the product... create a lineage of prints or refine a particular ... blah blah blah---

INVENTION TITLE

Methods and Compositions Relating To Extensible Transgenic Vector Assembler, Pestilence Ridder, Plus Cannabinoid Producer

DESCRIPTION

[Para 1] The present invention relates the compositions and methods for controlling harmful pestilence in toilets and sewage, using modular extensible genetic techniques, and the facilitation of rapid, low-cost, cannabinoid production.

[Para 2] Cannabinoids, traditionally synthesized in Cannabis Sativa, have served as agricultural reagents throughout the world for thousands of years. In light of recent technological advances one skilled in the arts may now synthesize these handy molecules in transgenic vectors such as e. coli and tobacco. This development may enable those skilled in the arts to supply enormous quantities of cannabinoid enriched media to consumers for the purpose of fungicidal, anti-microbial, and insecticidal use.

[Para 3] With regards to the issue of treating filth dispersed deep in underground sewers and inaccessible areas - the invention makes a stark contrast to previous arts. Whereas most chemical reactions must obey the laws of Brownian motion or undergo energetically unfavorable processes such as pumping (USPTO 5,360,556) or heating (USPTO 6,753,536), enzymatic reactions enabled in motile vectors hold a decisive advantage as they can move through a liquid medium more easily. As one skilled in the arts appreciates the possibility of using a motile plant vector such as the sperm of the gingko would allow even greater motility for the vector. In the preferred embodiment of this invention catalysts hosted in transgenic e. coli, transgenic tobacco root hair, and used in modular extensible vectors controlling the synthesis of compounds such as tetrahydrocannabinolic acid (THCA), cannabigerolic acid (CBGA), cannabichromenic acid (CBMA), the associated long term costs of pestilence control may be reduced dramatically - while simultaneously enriching the soil with valuable nutrients for commercial crops. As one skilled in the arts will appreciate - the long term application of the proposed invention will manifest itself in stages - much as any great culture ranging from ancient cheese and yogurt cultures to present day bio-engineered vectors, each application of the invention may, in the spirit of evolution, lead to a unique bio-transformation specifically adapted to its environment. The proposed invention brings to the table a base level of safer transmutation of certain toxic fungi (Llewellyn 1977), (Turner 1981), infectious microbes, (Van Klingeren 1976), (Schmitz 1973), and insect pests, (Quaghebeur, 1981) as well as infectious disease transmitted through insects such as West Nile Virus (McPartland, 1993). The nature of this invention is energetically favorable, easily propagated, and low up-keep in cost making it also ideal for third-world implementation in the pursuit of cleaner, safer land. In cases of emergency the preferred embodiment might also serve as a possible source of the neuroprotectant delta-nine tetrahydrocannabinol (THC) through the application of heat such as sunlight or direct flame. In the event of a terrorist attack of neurotoxins, for instance, one might as a means of last resort set fire to the growth medium to convert THCA to THC - which upon inhaling provides neuroprotection (Hampson 1998) (Van der Stelt 2001) (Mechoulam 2001) (USPTO 6,630,507). Whereas in prior arts Elsohy et al (USPTO 6,730,519) disclosed a method for reduced cost THC production they also rely on traditional abiotic, inorganic, energetically unfavorable means for THC extraction and purification of THC. Moreover their claims depend on natural growth of Cannabis Sativa, a process that may take up to fifteen weeks. Clearly this is not an acceptable waiting period in the case of a terrorist attack. In an alternate embodiment of the invention a serum of raw nutrients, as opposed to raw sewage, were used as the basic medium - in this case using modularized transgenic enzymatic techniques one skilled in the arts might produce several tons of THC in two to three days.

Description of Operation

[Para 4] As one skilled in the arts may appreciate the variety of vectors able to transform of the initial reagents (TSCSL, TOAP, TCAs, TTAs, TCBMs, TCBAs) into the desired reagents (THC, CBD, CBM) may result in hundreds or thousands of potential scenarios. Consider the heat that is generated in many compost conversion where temperatures may rise above 160 degrees Fahrenheit, in such cases it may be expedient to use an thermophilic vector, particularly for the incubation of the TSCSL, and TOAP. In the preferred embodiment of the invention it should be noted that the TTAs, TCBMs, TCBAs, are used in either a plant or animal vector - since cannabinoids exhibits both anti-microbial and anti-fungal activity it will require a non-microbial and non-fungal host.

[Para 5] In its preferred embodiment begin with a gigantic pile of refuse, that may include fecal matter, untreated sewage water, and decaying animal parts. It may be to the advantage of the user to initiate the enzymatic activity in a more sterile environment with nutrients needed for the synthesis of precursors of cannabinoids to alleviate environmental pressures of the sludge. In such cases as necessary the resulting enzymes and precursor products may be added directly to the filthy sludge or set aside and used as the growth medium for the transgenic cannabinoid synthesis with the resulting cannabinoids added to the filth sludge after their synthesis is completed. See FIGURE 1

[Para 6] Expose the pile of refuse to TSCSLs, TOAPs, and TCs teas - brewed as per the guideline in the literature commonly as anyone skilled in the arts will appreciate - and genetically modified to include promoters operably linked to bioluminescent proteins to help indicate and monitor effectiveness of the treatment. This tea is given from 24 hours to one month as indicated by bioluminescence (Figure 1) to finish blending in with the refuse - or as long as the bioluminescence appears active. (Figure 1. Step: Stilbene-like Synthase). These teas, when mixed with toxic bilge, enzymatically synthesize cannabinoids.

[Para 7] Next a root bed made of TTAs, TCBAs, and or TCBMs. Note the versatility of this invention. Any one of the aforementioned synthases, or indeed all three may be placed atop the pile bilge to create the desired reagents (ie THC, CBA, CBM). Also noteworthy is the elegant closed-loop nature of this system. By initiating the reaction with microbes that are not themselves immune to the final product the system will eventually turn itself off - as the reagent levels rise to higher levels the TSCSLs, TOAPs, and TCs die.

Description of Operation of Alternate Embodiments

[Para 8] The bucket containing the OAP is loosened atop a pile of crud, that may consist of any decaying or decayed matter, and that must consist of some decaying vegetable matter or living vegetation.

[Para 9] The bucket containing the CAS is loosened atop the pile of crud that previously received OAP treatment.

[Para 10] A blanket of roots from tobacco made of TTAs, TCBAs and or TCBMs are thrown over the crapulence and festering therein may it yield bountifully cannabinoids.

[Para 11] If time is of the essence one may speed up growth times by dispersing units of TEVs around the afflicted region through artificial or assisted means.

[Para 12] If precision in timing is desired it may be convenient to include a bioluminescent protein operably linked a functional promoter to the TEVs similar in methods to (USPTO 6,544,729) and created such as to reflect the activity of the TEVs.

Overview of Invention

[Para 13] This invention relates to the synthesis of cannabinoids for the purpose of general pestilence riddance in filthy organic and inorganic sludge. Through regulated enzymatic reactions, wherein cannabinoids with known anti-microbial, insecticidal, nematicidal, fungicidal properties and moreover nutritious, and neuroprotective, qualities are used to benefit regions where other commercial chemical reagents would require mechanized dispersion and cleanup. In plain English for those skilled in the arts - the genes involved in the enzymatic formulation of cannabinoids are inserted into foreign vectors thereby reproducing themselves and generating sufficient quantities of cannabinoids to clear the region of pestilence.

[Para 14] The advantages of this system are numerous. Whereas cannabinoid synthesis may not easily take place in Cannabis sativa due to its illegality, this invention is highly preferable. Whereas cannabinoid synthesis using inorganic techniques is not advantageous due to the inefficiency of inorganic and organic laboratory chemistry, this invention is highly preferable. Whereas most chemical synthesis routes for the creation of cannabinoids relate to the creation of extremely pure cannabinoids, this invention merely creates sufficient quantities as needed to rid a region of pestilence, and makes no claims whatsoever as to purity. Whereas the cost of creating cannabinoids synthetically would require large sums of money, as well as recurring costs for reagents, as well as a high degree of expertise and lab equipment, the invention described herein requires a single up-front cost to create the necessary vectors, and thereafter the invention may be distributed and applied to sludge and filth across the world with almost no requirements insofar a priori knowledge.

[Para 15] Using closed-loop modular enzymatic reactions, wherein each phase of catalysis may be halted by another counter reaction, and wherein each phase of catalysis may be easily monitored for effectiveness allows one skilled in the arts to more safely and effectively treat hazardous waste and the plethora of contagions therein. This invention refers to modular, in the sense that along the enzymatic pathway of choice each enzymatic building block is separated into a unique vector, uniquely identifiable by means bioluminescence and uniquely susceptible to a flavor of anti-microbial or anti-fungal such that the enzymatic process of choice may be halted at any given phase of production if desired. Modular may also or rather refer to the system as a whole, in that it should, handled by one skilled in the arts, leave little or no trace of enzymatically active reagent and be a closed-loop system - with the understanding that in nature there exists no such thing as an entirely closed-loop system, however, the preferred embodiment of this invention has in its design constructs a self-destruct or self-neutralizing mechanism for the living reagents. Thus in the preferred embodiment of the invention the catalysis of tetrahydrocannabinolic acid results in the recursive destruction of the initial vectors (Taura 2004).

Description of Objects

[Para 16] Polyketide Synthesis converts 3 Malonyl CoA plus 1 n-Hexanoyl-CoA to form OSCoA. This conversion may take place inside the muck and sludge, or may take place in a contained area and after the OSCoA.

[Para 17] Stilbene Carboxylate Synthase-Like (STCSL), in the case of Cannabis Sativa a Chalcone Synthase (CHS) that exhibits Stilbene Synthase (STS) activity in vivo and as per note in the literature (Raharjo 2004) there is reason to believe that the sequis used in the preferred embodiment of this invention and refers to any enzyme that generates 5-amylresorcinolic acid (olivetolic acid). While there are several enzymes capable of synthesizing olivetolic acid in the final analysis any enzyme capable of Olivetolic Acid synthesis will is sufficient. In the preferred embodiment the STCSL is inserted into the mitochondrial genome using the protofection technique (Khan 2004). The STCSL should be operably linked to bioluminescent protein to facilitate the monitoring of activity. The vector of the STCSL should also, in the preferred embodiment, have an operably linked In an alternate embodiment of this invention olivetolic acid is synthesized through inorganic techniques and thus added to the filthy sludge as a trigger molecule. In this manner the invention would have a limiting reagent from the offset, restricting the final output of pestilence ridders in such cases wherein limitations might be preferable. In another alternate embodiment of the invention the STCSL is chimeric with GOAP, or a pestilence ridding molecule.

[Para 18] Geranylpyrophosphate Olivetolic Acid Prenylase (GOAP) (Fellermeier 1998) is an integral part of this invention, and converts olivetolic acid into cannabigerolic acid. As one skilled in the art may appreciate any enzyme capable of yielding cannabigerolic acid is sufficient. In the preferred embodiment the GOAP is loaded into the vector in the manner described in Fellermeier’s work. In the preferred embodiment of this invention the GOAP is operably linked to a bioluminescent protein such as GFP or aequorin, and thus its activation is more easily monitored with minimal technical expertise. The GOAP is also operably linked to a promoter capable of up-regulating GOAP and thereby amplifying GOAP production.

[Para 19] Products made from these transgenic vectors should produce THCA, and, in addition, other precursor molecules as well as the necessary enzymes and proteins requisite for the aforementioned production, such as, tetrahydrocannibigerolic acid synthase, cannabigerolic acid synthase (CBGAS), cannabidiolic acid synthase (CBDAS), cannabichromenic acid synthase (CBRMAS), tetrahydrocannibinolic acid (THCA), olivetolic acid, polyketide synthase, and cannabigerolic acid synthase. Also disclosed is the unique and novel application of the TTAs in the function of a compost toilet additive and for the low-impact, sustainable, macrobiotic control of pests including Alabama argillacea (Riley 1885), Pieris brassicae (Beling 1932), Melolontha melolontha (Mateeva 1995), and Aphelenchoides composticola, (Grewal 1989).

The term "sequence identity" means that two polynucleotide sequences are identical (i.e., on a nucleotide-by-nucleotide basis) over the window of comparison. The term "percentage identical to a sequence" is calculated by comparing two optimally aligned sequences over the window of comparison, determining the number of positions at which the identical nucleic acid base (e.g., A, T, C, G, U, or I) occurs in both sequences to yield the number of matched positions, dividing the number of matched positions by the total number of positions in the window of comparison (i.e., the window size), and multiplying the result by 100 to yield the percentage of sequence identity. The terms "substantial identity" as used herein denotes a characteristic of a polynucleotide sequence, wherein the polynucleotide comprises a sequence that has at least 30 percent sequence identity, preferably at least 50 to 60 percent sequence identity, more usually at least 60 percent sequence identity as compared to a reference sequence over a comparison window of at least 20 nucleotide positions, frequently over a window of at least 25-50 nucleotides, wherein the percentage of sequence identity is calculated by comparing the reference sequence to the polynucleotide sequence which may include deletions or additions which total 20 percent or less of the reference sequence over the window of comparison. As applied to polypeptides, the term "substantial identity" means that two peptide sequences, when optimally aligned, such as by the programs GAP or BESTFIT using default gap weights, share at least 30 percent sequence identity, preferably at least 40 percent sequence identity, more preferably at least 50 percent sequence identity, and most preferably at least 60 percent sequence identity. Preferably, residue positions which are not identical differ by conservative amino acid substitutions. Conservative amino acid substitutions refer to the interchangeability of residues having similar side chains. For example, a group of amino acids having aliphatic side chains is glycine, alanine, valine, leucine, and isoleucine; a group of amino acids having aliphatic-hydroxyl side chains is serine and threonine; a group of amino acids having amide-containing side chains is asparagine and glutamine; a group of amino acids having aromatic side chains is phenylalanine, tyrosine, and tryptophan; a group of amino acids having basic side chains is lysine, arginine, and histidine; and a group of amino acids having sulfur-containing side chains is cysteine and methionine. Preferred conservative amino acids substitution groups are: valine-leucine-isoleucine, phenylalanine-tyrosine, lysine-arginine, alanine-valine, glutamic-aspartic, and asparagine-glutamine.

What is claimed is:

[Claim 1] Enzymatic reactions initiated in transgenic vector organisms generate cannabinoids - fungicidal, anti-microbial, and insecticidal in nature.

[Para 23] Regions of standing wastewater harboring high concentrations of unprocessed, unfiltered rubbage and manure can be natural sites for disease. The need for low-impact, low-maintenance composting solutions is needed to address city sewers and streets around the world that routinely overflow with toxic bilge. Controlling pests in such regions may often require the use of manufactured chemicals - created along costly and inefficient energy gradients (USPTO 5,227,537). Many of these chemical treatments present long-term hazards to the environment in the forms of run-off contamination, and build-up. Once introduced into a system these non-biodegradable inorganic compounds may not easily be eradicated. Although these methods may be suitable for certain bilge water, especially in treatment facilities where build-up and run-off are not concerns, they do not address the needs of farmers dealing with compost piles, nor sewage water running rampant through the streets of cities that have been obliterated by tsunamis, hurricanes, or tornadoes - wherein there may be an immediate need for rapid decomposition and pestilence ridding. Nor do these prior arts address the need for low impact conversion and or transmutation of toxins and pestilence, nor do these methods establish within the bilge an ecological breeding ground wherein one skilled in the arts might hope to use the nutrient rich, albeit highly toxic, solution for the establishment of biological byproduct. On the other hand, allowing the degradation of the toxic bilge to take place naturally may not be a viable option - as the aforementioned pestilence may soon make a bid to use the nutrient source as a home. In the proposed invention one skilled in the arts would safely apply numerous vectors to convert the bilge into valuable natural resources while simultaneously defending the region from harmful pestilence both through the creation of anti-microbial substances and through direct competition for resources - in much the same way that acidophilus in yogurt out-competes other microbes.

My kitchen now has two buckets with root in various stages of production, a third vessel carries the root in chunks, and another bucket contains the remnants of first round blending and grinding. Having weakened hands from weekend warrioring for the last several moons my grip on the machette has provided blisters well along my palms. The claims of kudzu as a hand cream and soap must be true - -- my silky-smooth blistery hands... oooh ahhhh.

Yup - that's right - Wednesday of last week Neptune stationed direct and we went live with the first draft of the Cannabinoid Biosynthesis Provisional Patnent - filed electronically via e-Pave 5.2 and linked together with the EFS-ABX package the whole thing rang in at a reasonable $100. Felt enormously fair - my favorite part was getting the graphics to load properly - kinda a chafe with the tif black and white specifications in place - 300 dpi is enough to have some mandelbrot fun - take a look at this sweet description from the drawings section

Well, that title says most of it. They're fighting things they don't understand, cause they're too chick-livered to fight things like hate, rape, violence etc... It's classic. And so now I live in a police state, America. Man o man o man. Whatta day. Whatta year. 20 billion or so according to the War on Drugs Clock jeez.... couldn't there be a more useful way to spend money? Fighting plants, fighting atoms, fighting things that occur naturally. Fighting the way people think by putting them in jail when they don't agree, or when their minds work differently from other people's. Prozac won't work on me. Neither will Ritalin. Cannabinoids mediate my central medial hypothalamus - an overactive part of my mind, and allow me some rest.

Marijuana and prayer go together like peas carrots and and black eyed peas. it's a known fact. Bob Marley preached it. That's almost passe now, though - the thought of fasting a week and smoking up and praying... yeah, that's right - giving thanks - peacefully, alone, silently - listening to God as the the world winds itself around It's Mighty Will. Whatever anyone else is talking about I'll never know, that moment of submission to Will is the only ecstacy worth mentioning, and yes, it can be enjoyed without cannabinoids, and yes, it can be enjoyed with them. It's personal, it's beyond words, and it's WELL BEYOND THE BOUNDS OF THE FEDERAL, STATE, AND LOCAL AUTHORITIES TO DICTATE! Those blind cowards. I do not hate them, I don't hate anyone. Hate of humanity is forbidden to me by my Master.

This is a project in the making - if anyone get's excited lemme hip you to this factoid - it's better for places where you really are SOL for power - out in the woods etc, and places like India, Sierra Leone, or wherever else... 100-200 Watts is a ton of juice if you wanna play a radio, and that's really still in theory mode for moi. My co-workers are well mannered, wholesome, and healthy. Whatta blast.

Still waiting on the Knoppix-GIS package to arrive from Sweden or wherever the heck. It will add a complete GIS solution to my desktop, and if I can find the seerver space for it I'll host it for you there... I downloaded v1.0- - it took about three days of periodic wireless hook-ups to get the 690 MB file. MD5'd it with fastsum and yup - there she is. Oh well, GRASS is a heck of a "solution" set - now it's time to get some data in there. I'm in the Charlottesville region, looking at working on a farm called Maple Hill farm - aka The best of what's around. So now it's time to figure out why anyone cares, or how it works - generally I don't even know enough to ask ask a question yet, so I'm asking the pro's who wrote the guidebook ANSWERS.

This is so cool -- GRASS started out as a US Military Project. They passed the torch on to Baylor and then the Italians (see above link) and Germans took the European lead picked it up from there. After that was done the download in Sweden (sourcepole link above). Git down with the global download / freenode. Ain't the net cool?

I'm working with the Corps of Cadets - the VMI reprobates - to finish up a massive database project that includes several million pages of documenation. Today I gave them a wholly unnecessary rally cry "We are fighting an enemy. Our enemy has a name. It's name is 'Stupid,' and it is everywhere. We will fight it with our data. We will send knowledge to the deepest darkest realms of the net, and we will win. We are getting ready to upload massive rounds of knowledge at mach speed. The scanner is your artillery. Load it with data, fire when ready."

I love giving pep talks, I think I'm getting psyched as much as anyone - plus the bonus affect is that I'm working with some really bad-ass dudes. GO VMI!!!

...and Allah Allah Allah.... it's hard to take the rest seriously. When in doubt, back to work, and the rest of the time extracting verbiage from a keyboard that might eventually synapse back --- in fact I'm counting on that. The machines have my vote. I've out-sourced so many neurons now it's starting to be a trend. whatever. I barely use them, and the best folks I know let them dance along strands of light - harmonic reasoning. Harmonic theory - aka super string theory - it's easy enough to understand if you think about it like a trampoline - two folks jumpin up and time it right and zzzzooooooooom yer way up there... or guitar strings - harmonic chords etc... so what's the matter - nehow - there it goes again askin questions - wonder which neuron ante'd that one up... quashed - it'll get its answer from Allah or not at all. tonight publishing data again - and fast. couple thousand pages to see what can be done if the computer knows everything about something other than girls and boys doing intimate acts. There's a new product out the called iMira that'll do the job that yer stupid parents can't gifure out how to implement, or your net-admin is to horny to get on....

soon though, soon - and it's not like popular is a thought, so yeah - wonderin' why South East Asia gets a Tsunami, California gets mud slides, and Florida gets Hurricanes? IT IS PURE COINCIDENCE, YA'LL :) Yup, sure guys - well in the mean time I got a bridge to sell - any bidders? vol 2 starts voy-la di dada... that's the simple answer. the long version has video and text too - like on treebay.org. I've stepped way outta line on that last rant on the indymedia sites - splurged and hinted at reality a few dozen times - dozin' now danzig tomorrow.
